Design and Layout of Microbiology Laboratory

A microbiology laboratory typically consists of five sections: a preparation room, a sterilization room, an aseptic room, a constant temperature incubation room, and a testing & analysis area.

1. Preparation Room (Sample Processing Area)

This room is mainly used for media preparation, sample pretreatment, and glassware cleaning. The recommended area is 20–30㎡. It needs to be physically isolated from but adjacent to the contaminated area, and located near the entrance to facilitate logistics. The room should be equipped with lab benches, fume hoods, centrifuges, reagent cabinets, sensor faucets, and eyewash stations, along with an independent drainage system to prevent backflow. The layout should ensure a unidirectional flow from preparation to sterilization, and then to aseptic operations.

2. Sterilization Room (Disinfection & Sterilization Area)

The core function here is the autoclaving and dry heat sterilization of culture media, instruments, and waste. The recommended area is 15–20㎡. It should be kept away from clean areas and close to the waste storage area. Main equipment includes at least two autoclaves (high-pressure steam sterilizers), a dry heat sterilization oven, and UV disinfection carts. The design requires a powerful exhaust system and dedicated exhaust ducts to prevent high-temperature steam from escaping; the flooring must be non-slip and heat-resistant.

3. Aseptic Room (Clean Operation Area)

Used for high-cleanliness operations like aseptic inoculation and strain purification, with a recommended area of 4–20㎡, typically maintaining a local Class 100 or Class 10,000 clean environment. Structurally, it must include a buffer room and an operation room, using a sealed color steel panel structure and epoxy resin flooring with no dead corners. Personnel must enter after changing clothes and passing through an air shower; doors are equipped with interlocking devices. The room maintains positive pressure to prevent contamination and is equipped with Class II biological safety cabinets, clean benches, and differential pressure gauges. UV light switches are located outside the room, and a constant temperature and humidity machine can be installed if conditions permit.

4. Constant Temperature Incubation Room (Microbial Culture Area)

Dedicated to the constant temperature incubation and observation of microorganisms, with a recommended area of 20–60㎡. It also adopts an inner and outer room design, with the outer room serving as a buffer and the inner room as the core culture area. A constant temperature and humidity air conditioning unit is required to control the temperature at 20–25℃ and humidity at 40%–70%. The room should avoid direct sunlight and vibration. Equipment includes various constant temperature/biochemical/mold incubators, shakers, and temperature monitors.

5. Testing & Analysis Area (Physicochemical & Molecular Testing)

This area handles tasks such as colony counting, PCR testing, and microscopic observation. The recommended area is 30–60㎡, and it must be independent of high-contamination areas. It is equipped with microscopes, microplate readers, PCR instruments, electrophoresis systems, and refrigerators with different temperature zones. This area has high electrical requirements, necessitating a UPS backup power supply. Lab benches should be vibration-proof and resistant to electromagnetic interference.
