Biosafety Laboratory Design and Construction

According to the international 'Laboratory - General Requirements for Biosafety' and 'Technical Specification for Biosafety Laboratory BuildingLevel four is higher. Generally, BSL-1, BSL-2, BSL-3, BSL-4 represent the corresponding level of biosafety laboratory; ABSL-1, ABSL-2, ABSL-3, ABSL-4 represent the corresponding level of animal biosafety experiments room.

Characteristics of different types of biosafety laboratories: a type: laboratories that operate normally considered non-airborne pathogenic biological agents; b1 type: can effectively use safety isolation devices (such as: biological safety cabinets) to operate Laboratories; Type b2: Laboratories that cannot be operated effectively with safety isolation devices.

The site selection and construction of the biosafety laboratory shall comply with the regulations and requirements of the national and local environmental protection and construction authorities.

The requirements for the location selection of laboratories of different levels in 'Technical Specifications for Biosafety Laboratory Buildings' are as follows:

Level 1 and Level 2: no requirement; Level 3: at least 20m away from public places and residential buildings. The distance between the building where the main laboratory is located and the adjacent buildings or structures should not be less than 1.2 times the height of the adjacent buildings or structures; Level 4: should be far away from the urban area. The distance between the building where the main laboratory is located and the adjacent buildings or structures should not be less than 1.5 times the height of the adjacent buildings or structures.

According to the above two national standards, the requirements for biosafety laboratory buildings at different levels are as follows:

Level 1: Shared buildings with controlled access doors for laboratories. There are windows on the door. There should be a sink at the exit. BSL-1 can have window, ABSL-1 should not have window.

Level 2: The building can be shared and communicated with other parts of the building, but a door with a lock that can be closed automatically shall be provided. There are conditions for storing spare items outside the work area. ABSL-2 should set up buffer room at the entrance and exit.

Level 3: The building can be shared with other laboratories, but it should form its own area. It should be located at one end or one side of the building, separated from other parts of the building by a closed door. The workshop in the protection area (in addition to the auxiliary work area) that directly engages in high-risk operations, the main laboratory, usually refers to the room where biological safety cabinets and animal isolators are located, also known as the core pharmaceutical cleanroom. Entering the main laboratory should pass through the buffer room; the intersection of the enclosure structure should be easy to clean and disinfect, and all gaps should be sealed; the door should be able to close automatically, with visible windows; there should be maintenance and cleaning space; Mandatory showers; there should also be buffer rooms at the entrances and exits of animal breeding rooms. Level 4: Independent building, or shared building with other levels of biosafety laboratories, but should be in a separate isolated area in the building; there should be a chemical shower; the enclosure of the protective area should be as far away from the building as possible Exterior walls; ABSL-4 shall have mandatory chemical showers.

Rooms with high risk of contamination in biosafety laboratories are usually equipped with biosafety cabinets, animal isolators and other equipment, called the main laboratory or core workshop. In the biosafety laboratory, the areas with high biological risk and requirements for airtightness and airflow direction are protected areas; areas outside the protected areas are auxiliary work areas.

Air purification measures of biosafety laboratory: 1. The first level of the biosafety laboratory can be naturally ventilated, and the second level can use an air-conditioning system with circulating air, which involves special operations and adopts full exhaust air. 2. The third- and fourth-level laboratory protection areas should adopt brand new wind. 3. The purification air-conditioning system should be equipped with three-stage air filtration at the fresh air outlet, the positive pressure section of the fan and the end of the air supply. Among them, the fresh air filtration can use a coarse-efficiency + medium-efficiency filter or a coarse-efficiency + medium-efficiency + medium-efficiency or sub-high-efficiency filter depending on the local atmospheric dust pollution. 4. The third- and fourth-level laboratories must be equipped with indoor air vents, and filters of national standard B (inclusive) or above should be installed near the air vents. At the same time, they should have the conditions for on-site in-situ scanning leak detection and disinfection. The four-level laboratory should have two exhaust high-efficiency filters. 5. It is advisable to adopt the upper and lower exhaust air flow organization, but the biosafety laboratory that raises large animals should adopt the upper and upper row method. 6. An airtight valve should be installed between the exhaust filter and the exhaust fan. 7. The exhaust should be opened before the supply, and then closed after the supply. The supply and exhaust are interlocked, and the exhaust fan should be backed up. 8. Maintaining the step-by-step negative pressure difference in the biosafety laboratory is the main task of air purification, ensuring that the airflow flows from the auxiliary area to the protection area.

The buffer room requires clean air supply and cleanliness. A positive pressure buffer room should be set up between the auxiliary work area and the outdoors.
